Oxygenation-Driven Radiobiology

During 1947–1962, research established tumor radiosensitivity as closely tied to tissue oxygenation, spurring strategies to reoxygenate tumors and tailor irradiation timing and dose. Concurrent advances in radiotherapy planning addressed tissue heterogeneity and precise dose estimation, while radiobiology matured with cellular survival modeling and organ-specific recovery concepts to sharpen dose–response understanding. Clinically, histology-informed planning began to guide treatment and adjunct approaches emerged to enhance efficacy.

Oxygen tension emerged as a central modulator of radiosensitivity, with studies showing higher tumor cell kill in well-oxygenated environments and proposing oxygenation strategies prior to and during irradiation to improve treatment efficacy [3], [4], [5].
